Инсулин синтезируется в β-клетках островков Лангерганса поджелудочной железы.

Как и многие секреторные белки, предшественник гормона (препроинсулин) содержит сигнальный пептид, который направляет пептидную цепь внутрь эндоплазматического ретикулума (см. Синтез белка и его созревание), где после отщепления сигнального пептида и замыкания дисульфидных мостиков образуется проинсулин. Последний поступает в аппарат Гольджи и депонируется в клеточных везикулах, β-гранулах. В этих гранулах путём отщепления C-пептида образуется зрелый инсулин, который сохраняется в форме цинксодержащего гексамера (см. Молекулярные модели: инсулин) вплоть до секреции.
