Обмен макромолекул, таких, как белки и РНК, между ядром и цитоплазмой осуществляется через ядерные поры (диаметр примерно 7 нм), образованные белковым комплексом. Поры регулируют транспорт через ядерные мембраны. Пептиды и небольшие белки, например гистоны, способны легко проникать в ядро. Более крупные белки (свыше 40 кДа) могут пройти через ядерную мембрану, только если они несут специфическую сигнальную последовательность.

Такая последовательность, ориентирующая белок на ядро, состоит из 4 основных аминокислот. В отличие от других сигнальных последовательностей, они не расщепляются при переносе белка в ядро. (см. Сортировка белков).
